Description:
CRISPR knock-in experiments require the presence of an HDR template (also known as a donor plasmid), which allows for homology-directed repair (HDR) to occur after Cas9 has cut the DNA forming a double-stranded break. The CRISPR HDR template consists of an insert sequence flanked by left and right homology arms that correspond to the sequence on either side of the CRISPR-induced double stranded break. For small mutations of <50 bp, we suggest using a ssDNA oligonucleotide template.
